Extraction, isolation, and expansion of tumour-infiltrating lymphocytes (TILs)

Identification of therapeutic targets by in silico genomic analysis in specific pathological conditions:

  • Transcriptomic or RNAseq databases from public or researcher-provided data will be used.
  • A study of the quality of the material will be carried out.
  • R-Bioconductor will be used for data normalisation and for the identification of differential expression between genes or conditions, or for the identification of genomic alterations of interest.
  • Analyses of functional enrichment, protein interaction, presence of the gene/protein under study in the surfaceome or association with prognosis can be performed, depending on the researcher’s needs.
  • The same analyses may be performed under specific conditions requested by the investigator, e. g., under treatment, drug resistance, or presence of a certain genomic (mutation, amplification) or epidemiological (age, smoking, or disease) condition.

Identification of biomarkers of activity or response: Using the same methodology described above, it will be possible to identify transcriptomic markers or genomic structural alterations associated with medical conditions, such as response or resistance to a given treatment.

Antigen design and antibody development (outsourced service):

  • Depending on the protein under study and based on its three-dimensional structure, the antigen of the most exposed part with antigenic capacity will be designed.
  • Depending on the protein under study, the antigen may be synthesised or produced after infection in mammalian cells (CHO or HEK293 cells).
  • Subsequently, immunisation and serum analysis, lymphocyte hybridisation, hybridoma screening and isotyping, oligoclonal hybridoma cloning and freezing of selected hybridomas, and antibody sequencing, production and purification will be performed.
  • Biochemical assessment of its specificity.

Pre-clinical evaluation of compounds-non-clinical pharmacology (TILs)

  • It can be offered as a service for different research groups that want to use these cells for cell co-culture studies under different pre-determined conditions.
  • Also, to evaluate markers of immune response as well as response to pharmacological compounds in in vitro experiments.

Cell culture techniques using cell lines and primary tissue:

  1. Genomic studies:
    • DNA and RNA extraction from cell lines.
    • siRNA silencing.
    • Quantification by quantitative PCR of selected genes, both at baseline and after interference treatments by siRNA silencing.
  1. Proteomic studies:
    • Protein extraction from cell lines at baseline and after treatment with different compounds.
    • Protein quantification using BCA, and subsequent absorbance reading in the spectrophotometer.
    • Evaluation by Western Blot of different proteins involved in different pathways related to Cell Cycle and Apoptosis.
    • Western blot evaluation of the target proteins inhibited by the compounds used. Assessment of drug effectiveness and toxicity.
